I am looking at showing groove a fillet weld preps on the same WPS. Can this done on an ASME WPS or are separate WPSs required?
I have seen this on WPSs, but that does not mean it's the correct thing to do.

Yes, this works fine. Sometimes different welder qualifications are required but not WPS

The WPS can become difficult to use if it becomes all too encompassing, but to include both groove and fillets on one WPS shouldn't be too difficult.

The most important thing to remember is the main reason for a WPS is to provide guidance to the welder.
As Al stated sometimes too much information is not beneficial - have to find a happy medium.
Hate to think how many times I have seen single WPS's with things like F, V, H, OH - with a range of 80 - 160 amps.
How can that assist anyone ?
