Does the vacuum situation have to be considered during designing a steam pressure vessel, which the vessel will be calculated with external pressure?

If the answer is yes, how to verify the stength of non-circular cross section subject to vacuum pressure?

Usually the pressure vessels containing steam, are designed for vacuum, due to the internal pressure drop during the steam condensation, with the temperature drop. That is, the vessel shall be designed for external pressure to a maximum of - 1 barg.
Vessels of noncircular shells are described in the ASME VIII Div.1 Appx. 13.

You have to read entire Appendix 13 referenced in the previous reply, to have an understanding of the design of noncircular vessels. The mentioned Appendix 13, under the clause 13.4 (f) is indicating the relevant design method for the noncircular heads. But first please read the Code.
