regarding the simple optimization problem you have...the best help you can get is from MATLAB help menu.Also check the site of Mathworks for several tips.
If you have specific queries...post a short description of your problem and i'd be glad to help you as far as i know...
Generally we use the gatool in matlab for optimazation.
