does " mass moment of inertia" of flexible body such as tire change during the motion?

Assuming the tire runs on soft ground without any deformation then goes on hard surface which cause deformation.
 
Yes, but not by much. If you are running a high frequency tire model such as Ftire it might just be detectable, at a guess.

Some drag slicks probably have a decent change in inertia as they grow with rpm.

I doubt there are many passenger car tires which change inertia by a significant amount during normal driving.
