when we are doing Hydro test activity, we are fixing the blind flange, to accelerate the pressure i need to install nozzle on the blind flange for this we have to go through any calculation ? and in which ASME standard should apply?

Look up ASME B 16.5 - table 6 and also see multiple posts here about holes in blind flanges. These are officially called reducing flanges.

Hole needs to be in the centre.

Depending on your pressure and flange rating, you might need to add a boss onto the flange to get sufficient thread depth.

Be very careful. Multiple incidents and injury has been caused by incorrect threaded test connection which are often subject to pulsing flow and forces from hoses and other connections.

My motto: Learn something new every day

Also: There's usually a good reason why everyone does it that way

when you say nozzle is understood as a welded nozzle, calculate for reinforcing as you do all other welded nozzles calculations including weld weld sizes.

logonkishore,
What size vent connection do you want to install on the cover of this Top Manhole?

Sometimes its possible to do all the right things and still get bad results

Easy way to avoid 'Flat Head' calc's; use an integrally reinforced fitting - O'Let - at the hole in your flange. Now your hole is fully reinforced; structurally that hole no longer exists.
