In installing a DMS sign, mounting holes were driven at the wrong location at a bottom truss. Would drilling new holes at the correct location and filling the unused holes with high strength bolts be an acceptable repair?

This is a question best asked of the engineer who designed the truss. It may be possible to drill the holes in the correct location and leave the incorrect holes blank, but it depends on many factors. Filling in the incorrect holes with bolts without consulting the engineer is negligent at best and may be considered deceptive.

For your instruction, please show me how "filling the unused holes" (in the beam) with bolts would restore the strength of the original beam's web or flange.

I can think of only one unusual set of circumstances where that idea "might" work.

FYI It is typical to 'stuff' misplaced boltholes with short bolts, for esthetics only. [and to keep Inspectors from asking why there are bolt holes w/o bolts]. But doing this w/o asking permission does smack of deception.

Built a sign truss for LaDOTD, and it was a DMS Truss. Their acronym meant Dynamic Messaging Sign, DMS.

Regardless of what DMS means for OP's job, the engineer on record needs to approve that "repair". Otherwise a plug will need to be put into the hole and welded in.
