I'm trying to spec out material to be used for a groundwater drilling rig tool and want to use 4140 steel but need to know whether to call out ASTM A322 and A29.

You could, if Commercial Quality Alloy is what you need and if Hot Wrought finish of raw material is suitable for your application.

An ASTM note remarks that:
"Some applications may require superior surface quality, or special chemical restrictions, metallurgical characteristics, heat treatment, or surface finishes which the purchaser may obtain by designating one or more of the available Supplementary Requirements."

You would specify ASTM A 322 not A 29. A 29 is a general spec.
