In order to have a vessel certified to ASME Code, it has to be manufactured by a U stamp holder, but dose the designer of the vessel have to ASME certified?

Spoonful, you don't specify but the reference to U Stamp implies Sec VIII, Div 1. Answer is no. I am not aware of any Sec VIII, Div 1 provision that addresses designer qualifications one way or the other.
Spoonful, even when meeting all relevant Code rules and employing the best engineering judgement the designer is always potentially liable for the design. As in any field.
Spoonful - from the perspective of the Code, there are no responsibilities for the designer (yet). All responsibility rests on the manufacturer. If the manufacturer contracts out design services, then there may be contractual obligations.
Spoonful, ultimate responsibilty still rests with the manufacturer. However: As the party contracting out the fabrication it is incumbent upon the design entity to insure the fabricator is meeting his responsibilities.
From ASME's perspective, I believe that what SnTMan said is correct - the responsibility is with the manufacturer, the holder of the U-Stamp (or its current incarnation). Everything else is a commercial/contractual concern. Is it important, yes; is it a requirement of the Code, no.
