I am trying to understand this distribution direction pulldown within Area Member loading (see attachment). I am trying to represent a live load, so I am wondering if I should select perpendicular to A-B, Two Way or something else? Also, what should I select for a wind loading? I looked through some of the youtube videos and they selected two way in one of them.

It depends on how you are expecting your loads to be distributed. Two way means that members in both directions will attract their trib width in load. A-B and the like are all one way.

Which would be the most conservative loading?

That again depends on the situation. and the framing orientation.
