please explain depending upon ANSI pressure class, how to design a weld for pipe and weld neck flanges

This is not something that can be easily done in a forum. You need to have someone with whom you can consult on a local basis.

Review ASME B31 Piping Code that is most applicable to your design and ASME B16.5. As mentioned above, get some help with this.
