I have a similar case with a hole and 4x1/2" threads on a flat end.
My question is regarding the threaded hole, are they allowed in asme sec viii div 1 and if so what is the section that allow it

Review UG-34, UG-39 and UG-43(e).
 
If are using a standard blind flange, look in B16.5 for the max hole that does not require reinforcing.
