please if any one know what eurocode say about the changing the effective depth in the same layer of concrete , is it acceptable to use different cover for the same plate , how could we asume our calculation

Very rare would be the code that prohibites something feasible. For a start, the codes themselves change the mandated effective cover of the rebar, depending upon fire risk exposition.

In the case of Spain, the concrete code EHE 98 and CTE (Código Técnico de la Edificación) mandate what to me look very reasonable covers for concrete plates, as pertains to such fire risk.

In any case you may use any reasonable cover; if you go too deep EHE 98 mandates supplementary (weaker) reinforcement closer to the surface to forfeit spall; and in any case an analysis for crack width may say you where you are respect the convenience to go deeper. Further than that one should suspend whatever the thing as a load.
