Plan 53B with continuous source of nitrogen not rechargeable

please can we use seal plan 53B bladder connected to available nitrogen tie in in plant.
instead of charging nitrogen by bottles

No. The pre-charge pressure for the bladder is lower than the operating pressure once it is placed into service. It functions based on the fact that the bladder was compressed when the pressure was increased in service. As fluid is lost from the system, the bladder will expand to push more fluid into the system so that differential pressure is maintained across the seal faces. If you had a permanently connected supply of nitrogen pressure, the bladder would not be compressed and would not function properly to maintain the seal differential pressure. Why would you want to have a nitrogen supply connected to the bladder? If you have leaks and are losing pressure, you should find and fix the leaks.

I may have misunderstood the question. You could connect the bladder to your plant nitrogen system and pressurize it to the pre-charge pressure. Then, you would block in the nitrogen supply and pressurize the barrier fluid side of the system. You would need to isolate the nitrogen supply away from the bladder before you pressurize the barrier fluid or you would have the problem I described above. Any additional charge added to the bladder would need to be made with the pump shut down and the barrier fluid depressured.
