I have acquiring an analog signal (sine wave) from an angle sensor measuring the clockwise and anticlockwise angular position.

By counting the periods of sine wave, I am able to measure the angular position. Now how I can get this information when direction changes?

Sorry you can not.

There is no way to know if whatever was rotating instantly reversed or not.

You would need a quadrature encoder which would show which direction the rotation is in.
