How do I stop Autocad from trying to read an empty drive a: every time I start it ?

Make sure one of your search path settings in Options doesn't reference A:\.

Probably what happened is that you opened a drawing in drive A on start-up, now it always wants to search for drive A first.  Try starting ACAD, and choose a template to start-up with, and then after it loads up, restart ACAD.  If that doesn't help, change your "rememberfolders" variable.
