I am designing a tapered spindle and I don't know what angle, length, etc. to use.
  20 000 rpm
  20 lb side load
  1 lb end load
  approximately 2 inch maximum tool lenght
  will operate 10 minute intervals, 2 times per hour, 10 hours per day
  want to use nut to hold tool in place and remove by hand - do not want to use a drift or draw bar to remove tool

Any guidance from tool designers?

Any matched taper will give you a lock. You will not be able to get the tool out "by hand" without some loving. As for design, the Handbook has all the standard tapers in it. Your question is a little confusing, if you design a tapered spindle that is none standard, are you also planning to design matching tool holders? A CAT 40 taper should be plenty good enough. If you're looking for something smaller let me know and I'll check on some other standards you could use.
