I see these two terms in marking ASME SA-480:

1- specification designation no.
2- material identification no

what is the differences and which one is equal Heat No.?

Neither, Those two are spec and grade. There is a longer list that includes identification of the mfg, the heat number, and a lot or process related ID.

The heat number is identified on the MTR under a separate heading or column.
