Does gasket design for Pipe flanges (ASME B16.5) and Orifice Flanges (ASME B-16.36) is same ?
Can I use ASME B-16.20 for Orifice flanges ??

Of course you can depending on the service, fluid, pressure and temperature that in the piping. Check your piping specification for the acceptable gasket.

In addition to saplanti's comments, make sure you specify the right pressure class for the gasket. Because of the additional flange thickness needed to accommodate the orifice taps, the orifice flanges may be a different class than the rest of the regular pipe flanges in the same system.
