I am working on a fatigue pressure vessel (Based on Div 2 critiria).
The question is that if the vessel can be designed, fabricated and registrated as per div 1 vessel and only add div 2 fatigue calculation on top of this? or the whole vessel has to be designed, fabricated and tested as per div2?
Is there any code to clarify this such as NBIC.

You can (and in my opinion should) design your vessel to Division 1, but apply the fatigue analysis per Division 2. In Div 1, UG-22 tells you to consider cyclic loading, but not how to do so. Then, you apply U-2(g) and go to Div 2 Part 5. Make sure to stay away from any partial-penetration welds, use as much NDE as you can afford, and make sure that you maintain the calculations and fabrication documents for the life of the vessel. It might be in your best interest to use a fracture-mechanics approach to determine your inspection interval. Despite your best efforts in design, you will likely have fatigue cracks, so you'll want to find them before they cause a problem.
