Is there a good reference manual for the proper placement and locating of HEPA filters in ceilings of clean rooms? And how strict is that placement/location?

IES-CC-RP-006 or 004 gives detailed reference. Installation should be perfect. You will get maximum leakage details also from that paper. Location depends upon where you want to use it. I can help if you have any specific questions.
