Can we weld a 6" diameter Pipe A53 grade B to 3/4" Steel Plate A36?

Yes, they are compatible materials for welding.

It is advisable though to check that the ratio of tube thickness to plate thickness be less than 1:3, otherwise some special design detail may be required to obtain a sound weld.
