Is there a GUI mode within Fluent6.0 to define geometry or I ve to rely on Gambit,Pro-E,etc softwares for my geometry.

hi !

as far as i know, fluent "only" does computation and post-processing....

But i worked on 5.5, not on 6.0   ....

Normally, i guess that Gambit comes usually with Fluent package...

Yep, Fluent needs a separate modelling program.
Gambit is commonly used however there are other possibilities.  You can generate your model in Pro Eng, Solid Edge, Ansys and others.  Try the fluent website.
