Does anyone knows the difference in the of application of Voltage differential protection (PTOV, 60) and Fuse failure supervision (RFUF)?

Its not same, differnt pinciple of operation and not only for REG670
function 60 is volatge phase differntial protection and two VT on the bus/line are needed.
FFS is voltage/curent function, operated on the U2/I2 or U0/IO or both of them principal.

REG670-function 60 does not.

Can't you just set it up to block the functions you want under signal matrix?

I'm not familiar with the REG670 to answer you.
I always saw 60 function blocking those functions 21.40, ...
I wonder what is the advantage of using 60 or FFS to block functions 40, 21, 27.

No advantages, its used, when you not have a second VT.
ActualLy, 60 is best solution.
Of course possible do it (if it prepared) in the single matrix.
