Hello all,
I have an assembly with multiple configurations, is there a way to configure the mates when you create them instead of having to go back after creating them and then RMB on the mate and selecting "configure feature"?  Basically, is there a way to specify what configurations the mate will apply to during the creation process?

Not during mate creation, but very easily afterwards using a Design Table.

You can also select multiple mates as part of the configure feature command.
