Hi All,

I am designing a 12,000 psi pump, incorporating a directional control valve. I am not sure on what type of high pressure seals I have to use. Do I have to use all of them example, O-rings, wipers, U- cups? Can someone please advise on a better idea?

What kind of pump is that? I suppose you have static and dynamic seals?

Look at examples of high pressure pumps and valve currently on the market.  O-rings and backup rings, metal-to-metal.

What are you pumping? flow rate?  Type of pump.

Assuming reciprocating water pump, look up high pressure washer pumps like Cat Pumps.  Should get some insights there.

As posted above by asimpson checkout the catalogue and find the size required by you.  Better yet if you know someone that has a pump your size order a seal kit and use same.  As long as you will make only one.
Early on we built our own high pressure pumps both for process and water blast cleaning.  We used Leather Vee or Chevron packing up to 2.5" diameter and in the 10,000 psig. range.

There are a usually a number of 10,000 psig pumps on the surplus market.
