Work for a manufacturing company that deals with a lot of subsea pressure equipment and was taking part in a pressure test for a cross over spool 13 5/8" 5k Flange x No 18 Hub (20 3/4" 3k). It was the No 18 Hub that was being tested which had a RX73 ring groove.

For the pressure test we went to 4500PSI 5mins and 15mins. I was wondering what are the design calculations needed in designing spools?

The piping code specification will give the design requirements.  Once you know what they are, then you can think about what calculations are required.  The basic code requirements are to ensure there is sufficient wall thickness, and determining that longitudinal stress of axial tension and compression plus bending stress are within allowable stresses under all design loading conditions.
