For automatic on-off valve(ball valve), should I test seat leakage according to standard ANSI FCI 70-2(similar with Globe control valve) or API-598 (similar to all type manual valves)

For on-off ball valves, use ASME B16.34 or API-598.  Only case I can think of for using FCI 70-2 (control valves) is if you happen to have a metal seated, characterized insert or other specialty ball valve that is designed to be used as a control valve.

Valve should be tested by API-598 as it is a block valve.

I agree with bcd and bj45.  FCI 70-2 publishes leakage rate classifications because control valves are expected to leak.  On/off isolation valves are expected not to leak.

The leakage rate for metal seated ball valves within API STD 598 is similar to ANSI/FCI 70-2.  API 598 requires zero bubbles per minute seat leakage for resilient seated isolation valves.  Some manufacturers of triple offset butterfly valves offer the resilient seated specifications.
