what is the regulation regard the raduis of unocupied area required around an enclosed packaged WWTP?

There are probably no strict regulations on siting wastewater treatment plants.

Metcalf & Eddy has suggested minimum buffer distances of 500 feet for aeration tanks and 1,000 feet for sludge holding tanks.

Check with the local authority who would review and approve the installation.  Most of them tell what the "setbacks" must be for various appurtenances.
