Pardon my lack of knowledge on the subject, but that's why I'm here.
Doing a retrofit job at a wastewater facility where the exist. system is cement lined ductile iron pipe with a combination of mech. jt. fittings and Victaulic fittings.
Question, when making a tie-in to an exist. 12" D.I.P. with
a flanged tee, how is the mating joint made?
1. by threading the exist. pipe in place and installing a mating flange?
2. via a new flg. spool and a grooved cplg?

A lot of times this is done via what is typically called a Uniflange.

Several manufacturers produce a Uniflange.  It is a flange that slides over the existing pipe and uses set screws to hold it in place on the pipe.

Do a web search on Uniflange and you will find it.

A mechanical fitting will work just fine.

1. do not thread the pipe in place
2.  I would also recommended the use of a uniflange flange adapter
3.  you could also use a piece of flange by plain end pipe and slip a bell of straight pipe over the plain end if it is buried.  Just another option.

The uniflange is your best option if the pipe is not under pressure.  If the line is under pressure use a Megaflange instead.  No concrete thrust blocks.  See this link. http://www.ebaa.com/Products/1000-2100/
