Does ASME III require the design specification to be certified first before material manufacture can start. or is required before fabrication of the vessel by the N-certificate holder ?

The owner's certified DS is required to initiate ANY work, even a quote, otherwise the vendor is designing blind and is therefore at their own peril.
In Canada you need the certified DS to even preliminary register the design with the authority.

Bottomline: The DS is worth bubkus unless its certified.
