What checks must I do to see if a wideflange beam can have a hole in it?

I do the following checks: Am I on the right track?

1-Check to see if how much section modulus was removed by the hole

2-Check to see how much I was removed

3-Check to see if shear is ok for section with hole.

You are on the right track.

I would try to locate the hole in the midle third of the span (assuming it is uniformly loaded) where the shear is relatively low. With such a shallow beam (10"), try to keep the center of the hole at the mid depth of the beam.

The procedure is a little complicated today so I suggest that you get a copy of AISC Steel Design Guide 2, "Steel and Composite Beams with Web Openings".

Yes - Design Guide 2 - there are a lot more than your three checks you list.  Perhaps 12  or so.

There is software out there that will check it for you.  RAM Beam is one of them.  If you have these quite a bit, the software is $400.  You could download the demo for free to try it.  If you like it, buy it.  Like I said, if you think you will have quite a few of these in the future, might be worth it.
