I'm going to use joint B-U4a(SMAW)as per AWS D1.1-2004.

The material specification are as follows;
ASTM A252 Gr.3,  thickness: 12.5mm
Single-bevel-groove weld, ? = 45°, R = +1/16, –0
with backing.

In this case, Whether Welding Qualification Test is required or not?

The value for root opening you provided, was the "as detailed" tolerance. For the groove angle you have selected, the specified root opening without tolerances applied is 1/4 in (6mm). The "as detailed" tolerance will permit you to specify it up to 1/4 in + 1/16 in = 5/16 in...and no lower. During fit-up, the "as fit-up" tolerances kick in.

The base metal specification does not seem to be covered in D1.1. Check Tables 3.1 and 3.2. If that is the case, then qualification testing in accordance with Section 4 will be required.
