I have many configurations of an assembly. When I add a part to the assembly and mate it, the new mates are suppressed in all configurations except the current one.

I know I can edit the mate properties after the fact, but can I change SW default to unsuppress new mates globally to all configurations?

Go to the Configurations tab.  Right-click on a configuration and choose "Properties".  On the resulting property manager there is a check box for "Supress new features and mates".  Un-check this box for each configuration.  By default when you create a new main configuration that box is checked.  It is un-checked by default when you create a derived configuration.
