Can a spacer be placed between two jam nuts and the jam nuts still function properly (not loosen)?

Sure.  They will probably work even better because you'll have some stretch beween the nuts.

If you put a spacer between two jam nuts, you have the equivalent of a regular joint with preload.

I'm with Cory Pad.  The Jam Nut Design
could have just as easily incorporated
a c'bore on the interface sides to
do what the spacer design would effect.
The thread designs in the standard jam
nuts are to be back to back.  That is
their strength and design purpose.
There was discussion earlier as to
whether they should be inboard or
outboard.  You might want to check out
that posting.

Jam nuts, with or w/o a spacer do NOT give a preloaded connection unless the bolt/stud is actually preloaded between the first nut and whatever anchors the bolt.
