I am trying to find out if anyone has had luck with drilling a 45 degree hole in the top of the stuffing box that comes out at the back of the impeller on a horizontal barrel pump? If someone knows of this and has a procedure for it I would appreciate the information.

Yeah, we have used a jig to drill a 15 degree incline vent hole inside 9/16 inch bore.  Obviously the drill ingress was close to the front of the box, thus allowing sufficient clearance for the holder.

I would try the same approach, make yourself a jig with that hole used as a pilot, then securely assemble it so that it would match the desired location of the drill.
