I am constructing a resonance box for my 4-cyl twin Weber car.Do I need to connect  the float chambers with the box or can they stay under atmospheric presure?

Assuming you're talking about DCOE's, They will need to be at the same pressure as the air horns.  That's what the hole in the face is for.  Just punch through your face plate and all the air signals will relate.
